ABSTRACT

BACKGROUND Leishmaniases encompass a spectrum of neglected diseases caused by parasites of the genus Leishmania, grouped in two forms: tegumentary and visceral leishmaniasis. OBJECTIVES In this study, we propose Friend Virus B NIH Jackson (FVB/NJ) mouse strain as a new experimental model of infection with Leishmania (Leishmania) amazonensis, the second most prevalent agent of tegumentary leishmaniasis in Brazil. METHODS AND FINDINGS We performed in vitro infections of FVB/NJ macrophages and compared them with BALB/c macrophages, showing that BALB/c cells have higher infection percentages and a higher number of amastigotes/cell. Phagocytosis assays indicated that BALB/c and FVB/NJ macrophages have similar capacity to uptake parasites after 5 min incubations. We also investigated promastigotes' resistance to sera from FVB/NJ and BALB/c and observed no difference between the two sera, even though FVB/NJ has a deficiency in complement components. Finally, we subcutaneously infected FVB/NJ and BALB/c mice with 2 × 106 parasites expressing luciferase. Analysis of lesion development for 12 weeks showed that FVB/NJ and BALB/c mice have similar lesion profiles and parasite burdens. MAIN CONCLUSIONS This work characterises for the first time the FVB/NJ mouse as a new model for tegumentary leishmaniasis caused by Leishmania (L.) amazonensis.

ABSTRACT

The purpose of this study was to characterize the genetic contribution to endothelial adaptation to exercise training. Vasoreactivity was assessed in aortas from four inbred mouse strains (129S1, B6, NON, and SJL) after 4 weeks of moderate intensity continuous exercise training (MOD), high intensity interval training (HIT) or in sedentary controls (SED). Intrinsic variations in endothelium-dependent vasorelaxation (EDR) to acetylcholine (ACh) as well as vasocontractile responses were observed across SED groups. For responses to exercise training, there was a significant interaction between mouse strain and training intensity on EDR. Exercise training had no effect on EDR in aortas from 129S1 and B6 mice. In NON, EDR was improved in aortas from MOD and HIT compared with respective SED, accompanied by diminished responses to PE in those groups. Interestingly, EDR was impaired in aorta from SJL HIT compared with SED. The transcriptional activation of endothelial genes was also influenced by the interaction between mouse strain and training intensity. The number of genes altered by HIT was greater than MOD, and there was little overlap between genes altered by HIT and MOD. HIT was associated with gene pathways for inflammatory responses. NON MOD genes showed enrichment for vessel growth pathways. These findings indicate that exercise training has non-uniform effects on endothelial function and transcriptional activation of endothelial genes depending on the interaction between genetic background and training intensity.

ABSTRACT

Collaborative Cross mice ( CC mice) are series of inbred mice strains generated from hybrid strains of mice with different genetic background which used for human complex diseases and genetic diversity diseases studies. Genetic diversity of CC mice can reflect different mouse subspecies, the single nucleotide polymorphism is four times than traditional inbred mice. CC mice are more and more widely used in the field of life science and medical research. Based on information retrieval of CC mice, we introduced the related information resources of CC mice origin, database, application tools, and research results, to promote CC mice resources to be used widely in China.

ABSTRACT

Objective To analyze the growth phenotype and blood biochemical parameters of chromosome 1 substi-tution mouse strain(CSS1), and investigate their potential of QTL mapping .Methods Body weight, body length, tail length, organ weight of the CCS1 mice were measured at different days to create a growth curve while blood biochemical in -dexes were measured at about the 80th day.Results The CCS1 mice were different from C57BL/6 mice in several inde-xes.Compared with the C57BL/6 mice during different developmental stages , six strains including B6-Chr1KM mice were significantly different in body weight .There were five strains including B6-Chr1CM mice significantly different with C57BL/6 mice in body length, and all of the CSS1 mice were significantly different from C57BL/6 mice in tail length.Part of CCS1 mice were significantly different from C57BL/6 mice in the weight of liver, spleen, kidney and brain.The ALT of female B6-Chr1CM mice was significantly higher than that in the C 57BL/6 mice.The ALP of female B6-Chr1HZ mice was signifi-cantly higher than that in the male C57BL/6 and B6-Chr1KM mice, and was significantly lower than that in the C57BL/6 mice.The TB of male B6-Chr1CM, B6-Chr1SMX and B6-Chr1HZ mice was significantly higher than that of the C 57BL/6 mice.The TG of male B6-Chr1SMX mice and male B6-Chr1TW mice was significantly higher than that in the C 57BL/6 mice. Conclusions The phenotype of Chr1 CSS mice is quite different from commonly used inbred strain C 57BL/6 mice.CCS1 mice show great potential in QTL mapping for their characteristic growth phenotype and blood biochemical indexes .

ABSTRACT

This study investigated the trabecular and cortical bone microarchitecture of tibia in 14-week-old C3H/HeN, C57BL/6J and ICR mice using micro-computed tomography (micro-CT). Defined volumes of interest were scanned at a resolution of 17 micrometer (isotropic). The X-ray tube was set at photon energy of 50 kV, current of 200 microA, exposure time 1.2 sec, and a 0.5 mm-thick aluminium filter. For quantification of bone mineral density (BMD), the bone samples were scanned by micro-CT together with 2 calibration phantoms. The image slices were reconstructed using 3-dimensional CT analyzer software. C3H/HeN mice showed significantly higher levels of bone volume fraction, trabecular number and BMD, and lower levels of trabecular separation, structure model index and degree of anisotropy compared to C57BL/6J or ICR mice in trabecular bone area. So the C3H/HeN mouse appeared to be a good model animal for the study on the changes of trabecular bone with high trabecular bone mass.

ABSTRACT

Objective To assess the dynamic age-related hearing loss of inbred mouse C57 and BALB/c strains via distortion product otoacoustic emissions(DPOAE). Methods DPOAE levels were evaluated in C57 and BALB/c of 20,30,60,90,120,150,180 days old respectively,with the conditions that two pure tones,which stimulated the DPOAE,were at frequencies(f 1,f 2,f 1/f 2=1.22)and levels(L 1=70 dB SPL,L 2=65 dB SPL).Results DPOAE levels of C57 and BALB/c gradually decreased with age.From 60 days old,DPOAE levels of the two strains were dramatically decreased at 4.0,6.0,8.0 kHz( P
